چکیده

Lung cancer is the leading cause of cancer-related deaths worldwide; non-small cell lung cancer (NSCLC), which includes squamous cell lung carcinoma, adenocarcinoma, large-cell lung carcinoma, accounts for more than 80% lung malignancy (Jemal et al., 2007). Despite recent advances in multimodality therapies, the prognosis for most patients with NSCLC is still unsatisfactory (Pfister et al., 2004). Recently, progress in the understanding of oncogenic kinase signaling pathways has provided better targets for developing effective therapeutic strategies, which may help to improve clinical outcomes. Mammalian target of rapamycin (mTOR) was first defined by Heitman et al. in 1991 and has attracted monumental scientific interest over the past few decade due to its broad involvement in many human cancers and other diseases (Heitman et al., 1991; Efeyan et al., 2010). mTOR is the master regulator of PI3K/Akt/mTOR pathway, and posses complex biological functions in the regulation of cell growth, proliferation, survival, as

Predictive Biomarkers and Personalized Medicine

Purpose:Non–small cell lung cancers (NSCLC) comprisemultiple distinct biologic groupswith different prognoses. For example, patients with epithelial-like tumors have a better prognosis and exhibit greater sensitivity to inhibitors of the epidermal growth factor receptor (EGFR) pathway than patients with mesenchymal-like tumors. Here, we test the hypothesis that epithelial-like NSCLCs can be dis...

Prognostic Significance of p27 Protein and Ki-67 Growth Fraction in Non-small Cell Lung Cancers

We immunohistochemically examined specimens of 215 surgically resected non-small cell lung cancers (NSCLCs) for p27 protein (p27) expression and the growth fraction determined by the Ki-67 labeling index (LI). The NSCLCs analyzed showed considerable heterogeneity in both p27 and Ki-67 LIs; 25 of 207 (13%) lacked p27 expression (p27 LI < 5%), and 116 of 215 (54%) showed a high Ki-67 LI (>30%). T...
